      目的:通过PPI网络分析确定CRC相关Hub基因,为CRC靶向治疗提供一定参考。方法:从OpenTargets数据库获得CRC相关基因,使用Metascape对基因集进行富集分析,确定基因集富集术语。采用STRING数据库及CytoScape构建PPI网络,采用MCODE寻找PPI网络中的功能模块(子网)。根据功能模块中节点的网络属性确定Hub基因,通过文献法和GEPIA数据库对Hub基因进行验证。结果:共得到CXCL8、ERBB2、CYCS 3个Hub基因,它们均与CRC的发生和发展有一定关系,。它们均在CRC组织与正常组织中存在差异表达,并与CRC患者的总体生存时间相关。结论:基于PPI的网络分析可准确预测CRC相关Hub基因和为CRC靶向治疗提供参考。
英文摘要:
      Objective To identify the colorectal cancer(CRC)-related Hub genes by PPI network analysis in order to provide reference for the targeted therapy of CRC. Methods CRC-related Hub genes were retrieved from OpenTargets and analyzed by enrichment analysis using Metascape to determine the enrichment terms of gene set. A PPI network was developed by STRING and CytoScape, in which the functional module (subnet) was identified. The Hub genes were identified according to the network properties of nodes in the functional module and verified by literature and GEPIA. Results CXCL8, ERBB2 and CYCS3 Hub genes were identified, which were related the occurrence and progression of CRC. The 3 Hub genes were differentially expressed in CRC tissues and normal tissues and were related with the overall survival time of CRC patients. Conclusion PPI-based network analysis can accurately predict the CRC-related Hub genes and provide reference for the targeted therapy of CRC.
